LONDON—As Prime Minister Boris Johnson prepares to lead the country out of the European Union, businesses are confronting the renewed prospect of leaving the bloc without a deal. This time, it is even more complicated.

Businesses have already spent millions of dollars stockpiling goods and making alternative transport arrangements to guard against potential disruption at the border. But for some, preparing for a divorce date in October is more challenging than it was for the originally scheduled exit in March.
